The play Much Ado About Nothing is one of the best plays I have read. It is sarcastic, exciting, and suspense filled. Shakespeare does a tremendous job of relating the characters to one another. The play has an awesome story line and a lot of humor to hold the audience's attention.


Much Ado About Nothing is about a man named Claudio who is the lord of Florence. Claudio wants to marry Hero, the daughter of Leonato, who is the governor of Messina. As this marriage is being arranged there are many sub-plots taking place. One of the sub-plots is that Hero has a cocky cousin named Beatrice who continually is fighting with, or mocking, Claudio's friend Benedick, a lord of Padua.


As the marriage draws nearer Don John, the bastard brother of Don Pedro, prince of Arragon, plots against Hero, for no reason, to make it look like Hero is a slut. Don John uses Margaret, Hero's gentlewomen, by having people misinterpret Margaret's sexual rendezvous as if she was Hero. There are also men that are run around and "watch and listen" and make trouble throughout the play. Their names are Dogberry, a constable, and Verges, a headborough.


Benedick finds out that Hero is supposedly a tramp and decides that he will not marry her, but he is going to wait to break it off. Benedick meets Hero at the alter with all their friends and family on their wedding day and tells everyone that Hero is a dirty whore and that he is not going to marry her. Hero passes out and Leonato, Hero's father, is furious. Meanwhile, Don Pedro, Claudio, and Leonato decide to try to get Benedick and Beatrice together. While Benedick is near they talk about how Beatrice loves him. Benedick reacts to this by writing her a love letter and she warms up to him also. They fall in love.


As a result of Hero being humiliated Friar Francis comes up with a plan. The plan is to pretend Hero died because of this hideous lie and to prove her virginity. Then they ask that Claudio marries her cousin to vindicate Hero. Claudio agrees and marries Hero's cousin, who is really Hero and all end well.


The climax is definitely at the wedding when Claudio tells everyone that Hero was not a virgin. It was a turning point and I knew something had to happen to turn things around vindicate Hero. It was a very good plan and it worked out for everyone.


One of the most memorable moments was at the party when Benedick and Beatrice were talking and making fun of each other. Another memorable moment is when Benedick is listening to Claudio, Leonato, and Don Pedro talk about how Beatrice likes him. I can just see him peering over and listening through the bushes.


I really liked the characters actions and responses to the other characters. This play was easy to read because it had so many interesting sub-plots going on at one time. There was also a lot of sarcasm going on and I love that. Now I absolutely have to go see the actual play!

As a 7 year old working adult I understand the importance of having an education. Working full time and taking care of your family makes it challenging to succeed in a higher education. You will experience a stronger drive to achieve if you build goals around what is important to you, like obtaining a higher level of education. I feel that a person really has to be focused in setting and achieving there personal goals. In my experience of trying to set personal goals in the past I found it to be very difficult to succeed at that. The reason for that was because I lacked the focus and the drive to be successful at that task.


One of my personal goals that I want to work on and achieve in as a University of Phoenix student is self-esteem. I've had low self-esteem basically all of my school life. During my early school days, it was difficult for me to focus at the task of learning. My so-called friends would tell me that I wasn't cool enough or the girls would tell me that I wasn't cute enough. It really tore my self-esteem down. One of my teachers once told me that "a positive self-esteem is both a prerequisite and a consequence of academic success." Of course I didn't know what he was talking about or what this even had to do with me. Now that I'm older and more mature, I do understand what he was talking about.


Another personal goal of mine is motivation. Having positive self-esteem helps a person to become motivated. With my low or negative self-esteem I also lacked motivation when I was in school. Since I wasn't cool or cute enough, I wasn't motivated to pursue my education. I want it to please others. I was motivated to please them, but not motivated to gain knowledge. Education at the time was not a priority of mine. With the atmosphere here at U.O.P, that lack of motivation will quickly turn into a rush of motivation.


Achievement is another personal goal. To achieve is to do successfully. I never achieved anything worth while in school, Because of the lack of a positive attitude and motivation. The word achievement also means accomplishment and the University of Phoenix has the resources to help me accomplish my goal. To graduate is my ultimate goal and to be able to have the opportunity to achieve that is truly a blessing.


One of my other personal goals that I want to achieve in is procrastination. People procrastinate because they don't believe in their ability to complete a task or don't believe in themselves in general. I procrastinated a lot in school. Assignments I had to do, I put of until the last minute and still didn't finish them in time. That's something that I want to achieve in breaking while I'm here at this University. It can hinder you in so many ways in your efforts to seek a higher education.


The second to last personal goal of mine is time management. Unlike high school, college requires students to spend much more time studying outside of class. In high school, I could get away with two-hour test preparation time or with writing a research paper in an hour or two. Not so in college. The main reason in managing time is to provide structure to my life and, in turn, piece of mind. So as my time management skills improve I feel that I would have less stressful situations resulting from procrastination here at the University of Phoenix.


My last personal goal is confidence. Confidence is trust, reliance and assurance. I had hardly any confidence in my ability to handle certain subjects which led to me procrastinating. For example, my weakest subject in school (and still is) is math. With me having no confidence no assurance that I can handle and overcome this task, it leads me to failing that subject. To gain confidence is my last personal goal that I want to achieve here at the University of Phoenix.


With my personal goals of achievement in order self-esteem, motivation, achievement, procrastination, time management, and confidence I ‘am ready to take on the challenge of being a student at the University of Phoenix. Th1 and Th cells are produced when DC1 and DC types dentritic cells present antigen to the TCR respectively (figure 1). The generation of an immune response both humoral by B cells and cell-mediated by Tc cells (CD8+) depends on the activation of Th cells. Th1 subset produces great amounts of IL- and IFN gamma; cytokines particularly important in cell mediated immunity. The Th subset is very good at providing B cell help by secreting IL-4,-5 and 6. The molecular basis for Th1- and Th-specific gene expression remains to be elucidated. However it has been found that Th cells express the GATA- gene whereas Th1 cells do not [].


The main role of the cytotoxic T cell, as the name suggests, is to kill other cells. The requirement for MHC class I on the target cell means that the Tc cell is very important in recognizing and destroying self-cells that have been altered or infected. The Tc cell must first become activated and mature into a cytotoxic T lymphocyte (CTL). These CTLs express on their surface molecules of a transmenbrane protein called the Fas Ligand. This is useful in ‘killing' most potential CTL targets as they express Fas (receptor for FasL) on their cell surface.


The T cell receptor made up of different heterodimers also contributes to phenotypic heterogeneity. The T cell receptors composed of and polypeptide chains can directly recognise antigens in the form of intact proteins or non-peptide compounds while those made up of and recognise antigens bound to MHC molecules. About 5% of peripheral blood T cells bear TCRs, most of which recognise non-peptide phosphorylated antigens []. TCRs and TCRs are both a result of rearranging gene segments. The chain is encoded by V, J and C segments. The chain is encoded by V, D, J and C gene segments. So rearrangement of the TCR and chain gene segments results in VJ joining for the chain and VDJ joining for the chain. Several mechanisms operate during TCR gene rearrangement and thus creating a high degree of diversity among T cell receptors. In my life, diversity plays a very important role.
